Opioids are highly effective analgesics that bind to mu receptors in the nociceptive pathway to reduce neuronal activity and pain perception. Mu receptors are also present in the limbic system where they produce reward, and as tolerance develops, patients are at risk of substance use disorders. 

Baseline drug testing should be performed prior to the initiation of treatment with opioids, followed by compliance monitoring during therapy.
